Expression Characteristics of Myofiber Types and Energy Metabolism Related Genes in Sika Deer
The number,size and type of muscle fiber are the basic characteristics of muscle fiber.The number and size of muscle fiber determine muscle yield,and the type of muscle fiber is closely related to muscle quality.In this study,the muscle tissues of sika deer of different sexes,including longissimus dorsi,quadriceps femoris,gluteus maximus and intercostal muscles,were used as experimental materials to study the following genes related to muscle energy metabolism.Paraffin sections of muscle tissues in different parts were made to detect and compare muscle fiber types,and the real-time fluorescent quantitative PCR technology was used to explore myofiber marker genes(MYH1,MYH2,MYH4,MYH7),mitochondrial activity related gene(ATP5F1A,PPARGC1A,PPARD),electron transport chain activity related gene(UQCRC2,SDHA,NDUFA9),glycolytic activity related gene(LDHA,LDHB,GK)in the muscle tissues of sika deer of different genders and different parts.The results showed that the cross-sectional area,diameter and density of muscle fibers of longissimus dorsi,intercostal and quadriceps femoris of male sika deer were higher than those of female sika deer,and the cross-sectional area,diameter and density of muscle fibers of gluteus maximus of male sika deer were lower than those of female sika deer.The expression level of the type Ⅱ A muscle fiber marker gene MYH2 was higher in the rib intercostal muscle,quadriceps femoris,and gluteus maximus of female sika deer.In contrast,the expression level of the type Ⅰmuscle fiber marker gene MYH7 was higher in the intercostal muscle and quadriceps femoris of male sika deer.Additionally,the expression level of type Ⅱ B muscle fiber marker genes MYH4 and MYH1 was higher in the intercostal muscle,longest back muscle,and gluteus maximus of female sika deer.The expression level of glycolysis related genes was higher in male sika deer,and the expression level of electron transport chain and mitochondrial activity related genes was higher in female sika deer.Hence,this study examined the significance of muscle fiber characteristics and associated gene expression levels in distinguishing muscle fibers in sika deer,aiming to provide valuable reference data for venison quality research.
