摘要

为研究梅花鹿成纤维细胞的分离培养方法并建立保种库,通过酶消化培养法和组织块培养法对梅花鹿原代成纤维细胞的分离培养效率进行分析.结果表明:酶消化培养法对梅花鹿成纤维细胞分离培养效果较好,并通过原代培养、冷冻和复苏建立了梅花鹿成纤维细胞保种库.研究结果为梅花鹿种质资源保存、体细胞核移植和基因克隆研究奠定基础.

Abstract

In order to study the isolation and culture method of sika deer fibroblasts and establish a seed bank,this study analyzed and compared the efficiency of isolation and culture of sika deer primary fibroblasts by enzyme digestion culture method and tissue block culture method,and proved that the isolation and culture of sika deer fibroblasts had achieved better results by the enzyme digestion culture method,and a sika deer fibroblast seed bank has been established through primary culture,freezing and recovery.The research aims to lay the foundation for the preservation of sika deer germplasm resources,somatic cell nuclear transplantation and gene cloning research.
