Effect of Melatonin on Sperm Survival of Cervus nippon in Vitro at Room Temperature
To study the effect of melatonin on the preservation of sika deer sperm at room temperature,fresh sika deer semen was divided into 5 groups.The control group was diluted with room temperature preservation diluent,and the 4 experimental groups were diluted with different amount of melatonin at equal multiples.The mass concentrations of melatonin in the diluted semen were 10,20,40,and 80 mg/L,respectively.Under normal temperature conditions,sperm motility,sperm motility,plasma membrane integrity,acrosome integrity,mitochondrial activity,and other indicators were detected every 4 hours until all sperm died,and the survival time and survival index of each group of sperm were calculated.The results showed that the addition of melatonin could reduce the survival time and survival index of sika deer sperm stored at room temperature,and there was a linear negative correlation with the addition amount of melatonin.Within 8 hours of storage at room temperature,when melatonin concentrations were 10,20 mg/L,the sperm motility and motility were significantly higher than those of the control group and the other experimental groups(P<0.05),and the 20 mg/L experimental group had the best effect.At 12 hours,the sperm motility of each experimental group was significantly lower than that of the control group(P<0.05).The sperm plasma membrane integrity rate showed a decreasing trend with time,no protective effect of melatonin on sperm plasma membrane integrity was observed.The addition concentration of melatonin within 20 mg/L did not have a significant effect on the sperm acrosome integrity rate(P>0.05),but the addition concentration of 40,80 mg/L significantly reduced the acrosome integrity rate(P<0.05).Within 12 hours,with the prolongation of time,the sperm acrosome integrity rate showed a decreasing trend,and the acrosome integrity rate of the experimental group was generally higher than that of the control group.When concentration of melatonin was 20 mg/L,the acrosome integrity rate was the highest.Within 12 hours,the mitochondrial activity of sika deer sperm increased first and then decreased.Adding 10,20,and 40 mg/L of melatonin significantly increased sperm mitochondrial activity compared to the control group(P<0.05).The addition of melatonin has a good effect on the storage of sika deer sperm at room temperature.The storage time should not exceed 8 hours,and the appropriate addition concentration is 20 mg/L.
