Changes of physiological and biochemical indexes during tissue culture seedlings rooting in Camellia oleifera
In order to determine therootingmechanism of tissue culture seedlinginCamellia oleiferaand promoterooting rate of tissue culture seedling, taking tissue culture seedlingsinC. oleiferaXL1asmaterials, changeregulations of nutrient substance, enzymeactivityand endogenous hormoneswere studied duringrooting. Theresults showed thatrootsin C. oleifera tissue culture seedlingwas derivedfrom the callus. During tissue culture seedlingsrooting, the content of soluble proteinswas decreased,while the content of soluble sugarwasincreasedifrstly, decreasedlater,and thenincreasedrapidly. PODactivitywasverylow duringinductionand expression ofroot primordium,while PODactivitywasincreasedrapidly duringinductionand expression ofadventitiousroots. PPOactivitywasifrstlyincreasedand then decreased. Additionally, the content of endogenous IAA, ZRand GAwereallincreasedifrstly, decreasedlater,and thenincreased,and they showed anincreasing tendencyasawhole,while the content of endogenous ABA showed"W" type of trend.
