Cloning and expression analysis of sucrose synthase gene (SS6) in Ziziphus jujube
Sucrose synthase is one of the key enzymes of sucrose metabolism,and has great significance in regulation of fruit quality and yield.In order to explore the detailed function of ZjSS6 in sucrose metabolic pathways of Ziziphus jujube,based on Z.jujube transcriptome data,using Z jujube ‘Zhongqiusucui’ as materials,and the full-length cDNA sequence of ZjSS6 gene was cloned by using RACE method,length of its coding sequences was 2 553 bp and it encoded 850 amino acids.Molecular mass and isoelectric point of the protein were 96.67 kDa and 7.57,respectively.Prediction results of protein structure showed that the protein coded by ZjSS6 contained two domain:sucrose synthase domain (SS domain) and glycosyl transferase group 1 domain (GT1 domain).The results of protein multiple sequence alignment and phylogenetic analysis showed that ZjSS6 had the closest relationship with the SS6 in Morus notabilis,and the next was in Prunus mume.The RT-PCR results indicated that ZjSS6 gene was expressed at all tissues (fruit,flower,shedding shoot,leaf and root),expression level was the highest in fruits at the young fruit stage,the next were in fruits and shedding shoots at the green fruit stage,but the overall expression level of ZjSS6 was trending downward during fruit development.
