Study on the rooting process and physiological and biochemical changes of the main branch cuttings of the Dendrocalamus latiflorus
[Objective]To explore the rooting process of Dendrocalamus latiflorus cuttings and its related physiological changes,and to provide a theoretical basis and practical guidance for the research of D.latiflorus cuttings propagation technology.[Method]The cuttings were treated with the growth regulator IBA and NAA in a 1:1 ratio at four concentration levels of 50,100,200 and 300 mg/L,respectively,with water as the control.The roots,rooting effect,rooting process and changes in endogenous hormones,oxidase activity and nutrients during the rooting process were measured to investigate the types of rooting and the physiological basis of rooting in D.latiflorus cuttings.[Result](1)The rooting effect of 200 mg/L IBA+NAA treatment on D.latiflorus branches was the best and significantly better than that of the control group;(2)To observe the morphological changes of the rooting process of the main branch of D.latiflorus bamboo,the rooting process could be divided into three stages:0-20 d for the root induction stage,21-60 d for the root elongation stage,and 60 d for the root maturation stage.Combined with the internal anatomical structure of the root system,the type of rooting of D.latiflorus cuttings belong to the submerged adventitious root primordial rooting type.(3)During the rooting process,the IAA content of the treated group was higher than that of the control group,both showing an increasing trend followed by a decreasing trend.The trend of ABA content in the treatment group increased before root maturity and then gradually decreased,while in the control group it decreased before 40 d and gradually increased after 40 d.The CTK content of the treated group started to decrease at 40 d and gradually increased after maturity,while the CTK content of the control group decreased during the elongation period and gradually increased after 40 d.The GA3 content of the treatment group decreased from 20-40 d and gradually increased after 40 d,while the GA3 content of the control group remained in a declining state until maturity.(4)SOD activity in the treated and control groups was highest at the root induction stage,then gradually declined and rose again after 80 d.POD activity increased after the induction stage,then gradually declined at 40 d,and stabilized at the root maturity stage,while POD activity in the control group was always at a lower level;IAAO activity gradually increased from root induction to elongation stage then slowly declined after 40 d.IAAO activity in the treated group was higher than that in the control group;PPO activity in both the treated and control groups tended to increase before the root maturity stage,then gradually declined after the maturity stage.(5)Both soluble sugars and soluble protein content changed during the cuttings,gradually decreasing before the root maturity stage and gradually increasing after the root maturity stage.[Conclusion]The IBA+NAA treatment has a significantly better effect on rooting of D.latiflorus cuttings than the control group.The joint coordination of endogenous hormones,oxidase activity and nutrients in the rooting process affects the formation and growth of adventitious roots of D.latiflorus cuttings,providing a theoretical basis and practical guidance for the study of D.latiflorus cutting propagation technology.
