[Objective]In order to provide a reference for root-limited cultivation of Lycium barbarum in the arid area of central Ningxia and promote ecological planting of L.barbarum.[Method]Taking four L.barbarum(four cultivars:Ningqi No.1,Ningqi No.7,Keqi 6081,Keqi 6082)that were cultivated for 3 years under the same root-limited cultivation conditions were selected as the research objects to explore the differences of soil nutrients in root zone among different cultivars under the root-limited cultivation mode,in the typical soil climate type area of Wangtuan town,Tongxin county,the central arid zone of Ningxia,and used Illumina HiSeq high-throughput technology and redundancy analysis method to study the community structure and diversity of soil bacteria and fungi in root zone,to analysis the relationships between soil nutrients and microbial community characteristics.[Result]Water content(WC),total nitrogen(TN),total phosphorus(TP)and total potassium(TK)contents of root zone soil of Keqi 6081 were the highest(except that the soil total nitrogen content of Keqi 6082 was slightly higher than that of Keqi 6081),which were 27.41%,37.41%,17.96%,2.31%higher than those of Ningqi No.1 soil,17.42%,24.69%,15.60%,and 4.53%higher than those of the Ningqi No.7 soil,and 16.93%,10.73%,3.49%higher than those of the Keqi 6082 soil,respectively;the soil organic carbon(SOC)content of the Keqi 6082 was the highest,which was significantly higher than that of the soil of Ningqi No.1,Ningqi No.7 and Keqi 6081 by 26.73%,5.82%and 14.20%,respectively;pH of soil of Ningqi No.1 and Keqi 6081 were significantly higher than that of Ningqi No.7 and Keqi 6082(P<0.05).The soil dominant bacterial phyla of the four L.barbarum cultivars under root-limited cultivation were basically the same.Among them,proteobacteria accounted for a larger proportion at the phylum level,and the abundance of the four cultivars soil was 35%or more,while the community composition of fungal genera was quite different,especially the soil fungi in the root zone of the Keqi 6081 showed a certain difference;Around the indicators that characterize soil nutrients,the other three cultivars were relatively scattered.Soil total nitrogen(TN),organic carbon(SOC),and total phosphorus(TP)were positively correlated with soil bacterial and fungal community composition and abundance.[Conclusion]Under root-limited cultivation conditions,there were differences in soil nutrient contents among four different cultivars of L.barbarum,which had no effect on bacterial community structure and diversity,but the fungi had great differences.Soil nutrients of Keqi 6081 and fungal community composition showed relatively large differences.The research results can provide reference for selection of excellent L.barbarum cultivars for ecological cultivation under the root-limited cultivation mode in the central arid zone of Ningxia.
