Effects of weed management methods on the soil bacterial communities and ecological function in plum orchard
[Objective]By analyzing the impact of different weed management methods on bacterial communities structure and ecological functions in plum orchards located in national key ecological county,the soil health management was provided.[Method]The soil bacterium DNA was extracted from plum orchards with natural-grass-covered,artificial-grass-covered,and herbicide treatments in different soil management and its adjacent secondary natural forest soils,then ITS genes were amplified,high-throughput gene sequencing and functional prediction were analyzed in bacterial diversity,communities structure and ecological functions.[Result]The number of OUT(operational taxonomic unit)in different treatments of soil ranged from 2012 to 3522,which belonged to 33 phyla,108 classes,242 orders,383 families and 667 genera.Chloroflexi,Actinobacteriota,Proteobacteria and Acidobacteriota were the main phylum,which above 80%of relative abundance in different treatments.The analyzing of bacterial diversity showed that Ace and Chao indexes of natural forest treatment were significant lower than 3 treatments of plum orchards.The analyzing of communities structure and different groups of soil bacterial showed that it was more abundance of Acidobacteriota,less disease bacteria,and a small abundance of Bdellovibrionia in natural forest soil.The disease related of Sphingomonadaceae was much more risk in plum orchards,then growing grass could to some extent increase the abundance of beneficial bacteria such as Bacillus,especially in natural-grass-covered.Artificial-grass-covered was beneficial for improving the diversity of bacteria in plum orchards,especially the enrichment of Gemmatimonadetes.The use of herbicide in plum orchard mainly enriched the species of Chloroflexi,but reduced the relative abundances of Bryobacteraceae and Bryobacter.Function prediction analysis showed that the most active was metabolism,mainly including carbohydrate metabolism and amino acid metabolism,involving chemoheterotrophy,cellulolysis,nitrogen_fixation and so on.Among all of treatments,artificial-grass-covered treatment had stronger nitrate reduction function,secondary natural forest treatment had stronger ligninolysis function,herbicide treatment had stronger methylotrophy function.[Conclusion]The Ace and Chao indexes of bacteria natural forest were significant lower than those of plum orchards,which was reflected in more abundance of Acidobacteriota.Different weed management methods had a significant impact on the plum orchard soil bacterial community structure and ecological function,and growing grass was beneficial for improving beneficial bacteria.
