Effects of 12C6+irradiation on seed germination and seedling growth of different varieties of Olea europaea
[Objective]To provide theoretical basis for irradiation mutagenesis breeding of Olea europaea.[Method]'Picholine','Chemlal','Arbequina','Koroneiki'and'Arbosana',five varieties of O.europaea seeds, were used as experimental materials and irradiated with five different doses of 12C6+(0,50,100,150 and 250 Gy),and the germination rate, survival rate, fresh mass, plant height, total leaf number, root length and lateral root number under different treatments after planting were measured. The effects of different radiation doses on seed germination and seedling growth of different varieties of O.europaea were evaluated according to subordinate function values analysis, and the semi-lethal dose was calculated.[Result]The effects of 12C6+irradiation on seed germination rate varied among the different varieties. With the increase of radiation dose, the germination rate of'Picholine'and'Chemlal'first increased and then decreased, while the germination rate of'Arbequina','Koroneiki'and'Arbosana'showed a continuously decrease trend.Exposed to 250 Gy,the germination rate was decreased to 11.46%,8.33%,14.58%,7.29% and 11.46%,respectively.12C6+irradiation inhibited seedling growth of different varieties of O.europaea.With the increase of radiation dose, fresh mass, plant height, total leaf number, root length and lateral root number were all decreased gradually, and all decreased to the lowest when treated by 250 Gy.12C6+irradiation had a certain lethal effect on O.europaea seedings, which was closely related to the irradiation dose.Compared with 40 d after germination, the survival rate of different varieties of O.europaea seedings on 90 d after germination was decreased in some different degree under different irradiation treatments. The total scores of the radiation doses of the five varieties were 0,50,100,150,250 Gy according to subordinate function values analysis, indicating that different irradiation doses(50-250 Gy)all affected the seeding growth of different varieties of O.europaea in varying degrees.Semi-lethal doses of five varieties of O.europaea seeds from high to low were'Arbequina'(153.7 Gy),'Picholine'(143.5 Gy),'Chemlal'(129.9 Gy),'Koroneiki'(115.3 Gy),'Arbosana'(108.1 Gy).[Conclusion]12C6+irradiation inhibits the seed germination and seedling growth of different varieties of O.europaea, and the greater the irradiation dose, the stronger the inhibition effect.Considering the indicators such as germination rate, survival rate, seedling growth, the recommended irradiation dose range of O.europaea seeds is 100 to 150 Gy.
