The effect of different intensities of UVA on the physiology and biochemistry of ginkgo hybrid offspring
[Objective]To explore the effects of different intensities of UVA on soluble sugars,soluble proteins,antioxidant enzyme activity,flavonoid metabolism-related enzyme activity,and total flavonoid content in ginkgo hybrid offspring,and to obtain the optimal light quality treatment to improve the total flavonoid content of Ginkgo biloba,providing a theoretical basis for the cultivation and breeding of ginkgo hybrid offspring.[Method]Using two-year ginkgo hybrid offspring seedlings as experimental materials,different intensities of ultraviolet light UVA(395-405 nm)treatment were set up.Under the same intensity of white light(300 μmol·m2·s-1),2,4,and 6 UVA ultraviolet lamps were added,with a UV intensity of 20 μmol·m-2·s-1(UVA-2),40 μmol·m-2·s-1(UVA-4),60 μmol·m-2·s-1(UVA-6).White light(300 μmol·m2·s-1)was used as the control(CK).[Result]Different intensities of UVA treatment had a significant impact on various physiological indicators of ginkgo hybrid offspring.With the increase of UVA intensity,the content of soluble sugars and soluble proteins showed a gradually increasing trend.The SOD,POD,PAL,C4H,and 4CL enzyme activities of golden-leaved G.biloba showed a trend of first increasing and then decreasing with the increase of UVA intensity.The SOD enzyme activity of green-leaved G.biloba increased with the increase of UVA intensity,while the POD,PAL,C4H,and 4CL enzyme activities showed a trend of first increasing and then decreasing.The total flavonoid content,DPPH,and ABTS free radical scavenging rate of ginkgo hybrid offspring showed a trend of first increasing and then decreasing,reaching their maximum after 20 days of UVA-4 treatment.The total flavonoid content of green-leaved G.biloba was the highest,at 28.65 mg·g-1,which was 56.3% higher than that of CK treatment.Except for SOD and POD,other physiological indicators of golden-leaved G.biloba were lower than those of green-leaved G.biloba.The correlation results showed that there were highly significant positive correlations between all physiological indicators of ginkgo.The multifactor ANOVA analysis revealed that leaf color,treatment time,and UVA intensity had highly significant effects on all physiological indexes of G.biloba,and the interaction effects of UVA intensity and treatment time had highly significant effects on antioxidant enzyme activity,flavonoid metabolism-related enzyme activity,flavonoid content,and DPPH and ABTS radical scavenging rate.[Conclusion]Different intensities of UVA treatment have a significant impact on various indicators of ginkgo hybrid offspring,with the highest total flavonoid content in green-leaved G.biloba treated with 40 μmol·m2·s-1 UVA for 20 days.
