Tissue culture propagation technology for 2 cultivars of Ziziphus jujuba
[Objective]To explore the effects of phytohormones on the induction,proliferation and rooting of adventitious buds in jujube stem segments,to establish the optimal hormone ratios of the medium at different growth stages,and set up a system for the efficient regeneration of jujube.[Method]Using the stem segments of the current year's young shoots of'Simusu'and'Fucuimi'as explant materials,the effects of different disinfection methods and time,types and concentration ratio of plant growth regulators on the induction,proliferation and rooting of adventitious buds,as well as the survival rate of transplanting were studied.[Result]The optimal disinfection system of the explant materials of two jujube varieties established in this study was 75% ethanol treatment for 30 s+2% NaClO solution treatment for 5-10 min.The optimal medium for inducing adventitious buds was MS+1.0 mg/L 6-BA+0.2 mg/L IBA+0.010 mg/L TDZ+30 g/L sucrose+5.6 g/L agar,the optimal proliferation medium was MS+1.0 mg/L 6-BA+0.2 mg/L IBA+0.006 mg/L TDZ+30 g/L sucrose+5.6 g/L agar,and the optimal rooting medium was 1/2 MS+0.1 mg/L IBA+0.1 mg/L NAA+0.3 g/L activated carbon+15 g/L sucrose+5.6 g/L agar.[Conclusion]The tissue culture system of two jujube varieties'Simusu'and'Fucuimi'with buds as explants was constructed.
