Research progress of oil-tea Camellia culture in vitro
In vitro culture of oil-tea Camellia is not only the technical foundation for rapid propagation,but also the technical foundation for biotechnology breeding.This article reviews in vitro culture of oil-tea Camellia buds and stem segments,as well as in vitro culture of embryos,cotyledons and leaves,and in vitro culture of anthers on the basis of the explant culture types.Special attention is paid to surface disinfection,browning,medium composition,and culture conditions.The use of buds,stem tips,and stem segments with buds is mainly for micropropagation.Although there have been many successful experimental reports,the factory rapid propagation using tissue culture technology has not yet been achieved due to difficulty in surface disinfection and browning problem of explants,especially difficulty in rooting.In vitro culture of embryos,cotyledons,leaves and other explants is most conducive to genetic transformation and gene editing,and has achieved varying degrees of success.However,due to the difficulties in culture and regeneration,there have been no reports of a stable transformation system based on this kind of culture.Anther culture is an effective method to obtain haploid plants,but it is currently difficult to regenerate plants.In short,oil-tea Camellia belongs to a type of recalcitrant plant species for in vitro culture.A review of the research progress on oil-tea Camellia tissue culture will provide useful references for researchers in its biotechnology.
