Objective To observe the effect of pterostilbene(PTE)in regulating autophagy to improve myocardial hypertro-phy in pressure overloaded rats.Methods Thirty SD rats were randomly divided into sham group,pressure overload group(abdominal aortic constriction[AAC]group),and AAC+PTE group,10 animals each group.Except for sham group,all other groups underwent AAC to establish a pressure overload myocardial hypertrophy model.PTE was treated by gavage,and saline gavage was given to sham group.Postoperative observation was performed for 4 weeks.Echocardiography was used to measure ventricular wall thickness and left ventricular fractional shortening(LVFS),Western blot was used to detect the ex-pression of autophagy-related proteins beclin1,microtubule-associated protein 1 light chain 3(LC3)and the expression of hypertrophic signaling molecule extracellular regulated protein kinases(ERK),and immunohistochemistry was used to de-tect the expression of oxidative stress index 8-OHdG.Results Ventricular wall thickness,beclin1,LC3-Ⅱ/Ⅰ,p-ERK,and 8-OHdG in the AAC group were higher than those in the sham group,while LVFS was lower than that in the sham group,and the differences were statistically significant(P<0.05).Ventricular wall thickness,beclin1,LC3-Ⅱ/Ⅰ,p-ERK,and 8-OHdG in the AAC+PTE group were lower than those in the AAC group,while LVFS was higher than that of the AAC group,and the differences were statistically significant(P<0.05).Conclusion PTE can ameliorate myocardial hypertrophy in ACC rats,and the mechanism may be relat-ed to the regulation of autophagy and reduction of oxidative stress.
