Analysis of the mechanism of Yiqi Tongluo Decoction in treating diabetic peripheral neuropathy based on network pharmacology
Objective To investigate the mechanism of Yiqi Tongluo Decoction in the treatment of diabetic peripheral neuropathy(DPN)by network pharmacology.Methods The core components of Yiqi Tongluo Decoction were screened and the target proteins were predicted by means of traditional Chinese medicine system pharmacology database and analysis platform,traditional Chinese medicine comprehensive database and traditional Chinese medicine syndrome association database.The related genes of DPN were searched by GeneCards,OMIM and TTD databases,and the target proteins of Yiqi Tongluo Decoction were screened by Wayne diagram.Construction of core component-target protein network diagram by Cytoscape.Drawing protein-protein interaction network of Yiqi Tongluo Decoction by String database.GO and KEGG enrichment analysis of target protein were carried out by using Metascape database.Results A total of 191 core components were screened out from 10 kinds of traditional Chinese medicines,involving 263 target proteins,of which 189 genes were related to DPN.The results of network analysis show that the biological processes involved in the treatment of DPN by Yiqi Tongluo Decoction are mainly concentrated in the negative regulation of cell population proliferation,DNA metabolism process and the regulation of protein hydrolysis.The molecular functions involved are mainly concentrated in lipid binding,kinase binding,protein kinase binding,etc.Cell components are mainly concentrated in lysosomes,axons and postsynapses.Its mechanism may be related to PI3K-Akt,MAPK,calcium,Ras,cAMP,Rap1,chemokines,NOD-like receptors,Wnt,cGMP-PKG,JAK-STAT,Hippo,mTOR,oxytocin,phospholipase D,regulating stem cell pluripotency,Apelin,estrogen,insulin and apoptosis pathway.Conclusion This study initially revealed the characteristics of Yiqi Tongluo Decoction's multi-target overall regulation of DPN,which provided new ideas and methods for further clinical research on the mechanism of this prescription in treating DPN.
