Effect of dioscorea porridge on insulin resistance and oxidative stress in rats with diabetes and stroke
Objective To investigate the effect of dioscorea porridge on insulin resistance and oxidative stress in rats with diabetes and stroke.Methods A total of 60 Wistar rats were selected,10 were used as blank group,and 45 rats were treated with abdominal injection of Streptozocin(STZ)and ZeaLonga thread thrombus.The rats with successfully constructed mem-branes were randomly divided into model group and dioscorea group,and 8 rats in blank group,12 rats in model group and 13 rats in dioscorea group.Model group and blank group were given 0.9%Sodium Chloride Injection 5 ml,and dioscorea group was given dioscorea porridge 5 ml,a total of 4 weeks.After the intervention,fasting blood glucose(FBG)was measured by portable glucose meter every 2 weeks,and oral glucose tolerance test(OGTT)was performed after the intervention for 4 weeks.Glycoylated hemoglobin(HBA1),fasting insulin(FINS)and homeostasis model assessment-insulin resistance(HOMA-IR)were detected by enzyme-related immunosorbent assay.The blood sugar of rats was detected,and the liver was taken to detect the oxidative stress markers superoxide dismutase(SOD)and malondialdehyde(MDA).The key proteins of insulin resistance signaling pathway(glucose transporter 4[GLUT-4],insulin receptor substrate 1[IRS-1],phosphorylated insulin receptor substrate 1[P-IRS-1]),and colon tissues of each group were taken 100 mg to detect the expression of glucagon-like peptide-1(GLP-1).Results Before the ex-periment,there were no statistical differences between the two groups(P>0.05).After the experiment,FBG,HBA1,FINS and HOMA-IR of 2 weeks and 4 weeks rats in dios-genin group were lower than those in model group,and the differences were statistically significant(P<0.05).The glucose tolerance of rats in diocorea group after OGTT was lower than that of model group,with statistical significance(P<0.05).After the experiment,GLP-1 in dioscorea group was higher than that in model group,with statistical significance(P<0.05).The key proteins of insulin resistance signaling pathway IRS-1,P-IRS-1 and GLUT-4 in dioscorea group were higher than those in model group,and the differences were statistically significant(P<0.05).The oxidative stress markers MDA in dioscorea group was lower than those in model group,SOD was higher than that in model group,and the differences were statistically significant(P<0.05).Conclusion Dioscorea porridge can effectively improve blood sugar,oxidative stress and IR in rats with diabetes and stroke,which is worthy of clinical application.
