Simultaneous determination of four immunosuppressive drug concentra-tions in serum by liquid chromatography-tandem mass spectrometry
Objective To establish a method for the determination of serum sirolimus,tacrolimus,everolimus and cy-closporine A.Methods Four immunosuppressants in serum were quantitatively detected by liquid chromatography-tandem mass spectrometry.The four immunosuppressants were treated with the isotopes sirolimus-d3,tacrolimus-d4,everolimus-d4 and cyclosporine A-d12 as internal standard.Results The four immunosuppressants showed a good linear relationship with the ratio of peak area in their respective linear ranges.The lower limit of quantitative measurement of the instrument met the requirements of clinical detection.The precision of the instrument within the batch and between the batch were within 15％,the accuracy deviation was within the range of±15％,and there was no obvious carrying contami-nation.Conclusion The method is simple,rapid,sensitive and specific,and can be used for the determination of four im-munosuppressants in serum.
