Optimization of quantitative determination method for the content of Ephedrine Hydrochloride and Pseudoephedrine Hydrochloride in Wuwei Ganlu Medicinal Bath Wash
Objective To accurately determine the total content of ephedrine hydrochloride and pseudoephedrine hy-drochloride in Wuwei Ganlu Medicinal Bath Wash,and to optimize sample pretreatment methods and liquid chromatographic conditions.Methods Accurately measuring 3 ml of the sample was entailed in the pre-treatment method,followed by ultra-sonic extraction with methanol for 45 minutes.After cooling,the solution was brought to a final volume of 25 ml.A precise amount of 1 ml of the filtrate was collected and placed on a neutral alumina column.Elution was carried out with 9 ml of 50%methanol,the eluate was colleted and adjusted to a final volume of 10 ml.Chromatographic conditions:the chromato-graphic column was Aglient Eclipse Plus,C18(250 mm × 4.6 mm,5 μ m),and the mobile phase was acetonitrile-0.2%phosphoric acid solution(3:97)and the injection volume was 20 μl.The wavelength was 210 nm.The flow rate was 1 ml/min.The column temperature was 30℃.Results The linear range of ephedrine hydrochloride was 7.232~28.928 μg/ml(r2=0.999 5,n=7),and the linear range of pseudoephedrine hydrochloride was 2.704~10.816 μg/ml(r2=0.999 2,n=7),the linear relationship was good,and the average sample recovery rate of the sum of the two contents was 100.33%,the relative stan-dard deviation(RSD)=0.3%(n=8).Conclusion This method can be used to determine ephedrine hydrochloride and pseu-doephedrine hydrochloride at the same time,which is easy to operate,accurate and reproducible,and can be used for quality control of Wuwei Ganlu Medicinal Bath Wash.Through the application of this method,it can ensure that the product meets the specified standards,ensure the quality of the product,and provide reliable technical support for related research and production.
