摘要

目的 探讨补肾清热方对大鼠牙槽骨吸收的影响及其潜在机制.方法 选取54只SD大鼠,按随机数字表法分为空白对照组、模型对照组、阳性对照组、实验组(A1、A2、A3)共6组,每组9只.除空白对照组外,其余五组均采用结扎左上颌第一磨牙+10%糖水饮用的方法诱导牙周炎大鼠模型.造模成功后,实验组(A1、A2、A3)分别用3.5、7、14ml/(kg·d)等剂量补肾清热方灌胃,4周后取各组SD大鼠左上颌第一磨牙及其牙周组织.检测各组大鼠的左上第一磨牙探诊深度(PD),并采用Micro CT检测左上颌第一磨牙釉牙骨质界(CEJ)-牙槽嵴顶(ABC)的距离,采用Western Blot检测骨保护素(OPG)、核因子-κB受体活化因子(RANK)、核因子-κB受体活化因子配体(RANKL)蛋白表达.结果 干预4周后,阳性对照组及实验组(A2、A3)的PD小于模型对照组,差异有统计学意义(P<0.05);实验组A1与其他组间PD比较,差异无统计学意义(P>0.05).Micro CT检测结果显示,模型对照组的CEJ-ABC距离大于空白对照组,差异有统计学意义(P<0.05);阳性对照组、实验组(A1、A2、A3)的CEJ-ABC距离均小于模型对照组,差异有统计学意义(P<0.05).Western Blot检测结果显示,模型对照组与阳性对照组的OPG表达水平均低于空白对照组,差异有统计学意义(P<0.05);其他各组间的OPG表达水平比较,差异无统计学意义(P>0.05).模型对照组的RANK及RANKL表达水平均高于空白对照组,阳性对照组、实验组(A1、A2、A3)的RANK及RANKL表达水平均低于模型对照组,实验组(A2、A3)的RANK及RANKL表达水平均低于阳性对照组,差异有统计学意义(P<0.05).结论 补肾清热方可抑制牙周炎大鼠牙槽骨的吸收,其潜在机制可能是通过OPG/RANK/RANKL信号系统参与.

Abstract

Objective To investigate the effect and mechanism of Bushen-Qingre Prescription on alveolar bone resorption in rats with periodontitis.Methods A total of 54 SD rats were selected and divided into 6 groups(blank control group,model control group,positive control group and experimental groups[A1,A2,A3])according to random number table method,with 9 rats in each group.Except for the blank control group,the periodontitis model was induced by ligating the left maxillary first molar and drinking 10%sugar water in the other five groups.After the model was successfully established,the rats in the experimental groups(A1,A2,A3)were treated with Bushen-Qingre Prescription(3.5,7,14 ml/[kg·d])by gavage,re-spectively.Four weeks later,the left maxillary first molar and periodontal tissues were collected.The probing depth(PD)of the left upper first molar was measured,and the distance between cement to enamel junction(CEJ)and alveolar bone crest(ABC)of the left upper first molar was measured by Micro CT.And the protein expression of osteoprotegerin(OPG),recep-tor activator of NF-κB(RANK),receptor activator of NF-κB ligand(RANKL)were detected by Western Blot.Results After 4 weeks intervention,the PD of positive control group and experimental groups(A2,A3)were less than that in the model control group,and the differences were statistically significant(P<0.05).There were no significant differences in PD between experimental group A1 and other groups(P>0.05).Micro CT detection results showed that the CEJ-ABC distance of model control group was greater than that of the blank control group,and the difference was statistically significant(P<0.05).The CEJ-ABC distance of the positive control group and the experimental groups(A1,A2,A3)were smaller than that of the model control group,and the differences were statistically significant(P<0.05).Western Blot test results showed that the expression level of OPG in model control group and positive control group were lower than that in the blank control group,and the differences were statistically significant(P<0.05).While there were no significant differences in OPG expression among other groups(P>0.05).The expression levels of RANK and RANKL in the model control group were higher than those in the blank control group,the expression levels of RANK and RANKL in the positive control group and the experimental groups(A1,A2,A3)were lower than those in the model control group,and the expression levels of RANK and RANKL in the experimental groups(A2,A3)were lower than those in the positive control group,and the differences were statistically significant(P<0.05).Conclusion Bushen-Qingre Prescription can inhibit alveolar bone absorption in periodontitis rats,and the potential mechanism may be involved in OPG/RANK/RANKL signaling system.
