Objective To establish a method for simultaneous determination of serum Citalopram and Mirtazapine in pa-tients with depressive episode by high performance liquid chromatography-mass spectrometry(LC-MS).Methods The chromatography was performed on Acquity UPLC BEH C18 column(2.1 mm × 50 mm,1.7 μm)with mobile phase consisting of acetonitrile-10 mmol/L ammonium acetate aqueous solution(all containing 0.1%formic acid)with gradient elution at a volume flow rate of 0.5 ml·min-1.The column temperature was 40℃.Electrospray ionization positive ion(ESI+)scanning,multi-reaction monitoring mode detection,retention time and ion abundance ratio qualitative,vardenafil as the internal standard quantitative.Results Citalopram and Mirtazapine showed a good linear relationship in their respective mass con-centration ranges,the correlation coefficient r2>0.99,the lower limit of quantitation was 2.5 ng/ml,and the relative standard deviation(RSD,n=6)was 1.5%-9.3%.Conclusion High performance liquid chromatography-mass spectrometry can si-multaneously determine the serum concentrations of Citalopram and Mirtazapine in patients with depression.The results are accurate and reliable,which is conducive to clinical rational drug use.
