Fermentation Optimization of Pestalotiopsis uvicola GMH31 against Botryosphaeria dothidea on Kiwifruit
Strain GMH31 was Pestalotiopsis uvicola isolated from Artemisia japonica,and it could control efficiently Botryosphaeria dothidea on kiwifruit.Antifungal active test was studied by poison medium technique.Single factor experiment and orthogonal experiment were adopted to optimize liquid fermentation media (carbon source,nitrogen source,inorganic ions and trace elements) and conditions (fermentation time,culture temperature,pH,rotating speed,medium capacity and inoculums amount).The results showed that the optimal compositions of cultural medium were as follows:maltose 35 g/L,NH4Cl 12 g/L,K2HPO4 0.1 g/L and glycine 0.015 g/L.The optimal fermentation conditions were as follows:fermentation time 14 d,cultured temperature 26 ℃,initial pH 6.0,rotating speed 120 r/min,medium capacity 100/250 mL,and inoculation rate 6%.Inhibitory rate of strain GMH31 fermentation filtrate against Botryosphaeria dothidea on kiwifruit increased by 16.45% under the optimal fermentation medium and culture conditions.
