农杆菌介导的玉米幼胚遗传转化体系的建立

Establishment of Genetic Transformation System of Maize Immature Embryos Mediated by Agrobacterium tumefaciens

采用玉米幼胚为受体材料,利用农杆菌介导植物表达载体pCAMBIA 1300-35S-GUS-Nos侵染玉米幼胚,以GUS基因瞬时表达率为指标,通过单因素试验及四因素三水平正交试验研究了基因型、幼胚大小、侵染菌液浓度及侵染时间对幼胚遗传转化率的影响.结果表明:各因素对幼胚遗传转化率影响的大小依次为基因型>幼胚大小>侵染菌液浓度>侵染时间;最佳侵染组合基因型为H99、幼胚为1.1~2.0 mm、侵染菌液OD600=0.6、侵染时间为30 rain,GUS基因对幼胚的瞬时表达率可达(34.44±1.11)%.
With maize immature embryos as receptor materials,the plant expression vector pCAM-BIA1300-35S-GUS-Nos was introduced into maize immature embryos by Agrobacterium-mediated method.With GUS transient expression efficiency as indicator,the factors,such as,genotype,immature embryo size,infection liquid concentration and infection time,were studied by single factor test and the L9(34) orthogonal experiment to explore their effects on genetic transformation rate.The results show that:The effect rank of these factors was genotype>embryo size>bacterial concentration >infection time;The optimal infection rate was the genotype H99,the size of immature embryos was 1.1-2.0 mm,OD600 =0.6,the time of infection was 30 min,and the transient expression rate of GUS gene was (34.44± 1.11) %.
