中华散尾鬼笔提取工艺响应面优化及其粗提物毒力测定

采用超声提取法,以超声时间、乙醇浓度、提取温度、料液比、超声功率5因素进行单因素提取试验,而后选择对中华散尾鬼笔粗提物提取率有显著影响的3因素进行响应面分析,优化出中华散尾鬼笔粗提物的最佳提取工艺。试验结果表明:在料液比1∶80、乙醇体积分数60%、超声时间80 min时,提取率最高,为31。21%;以舞毒蛾3龄幼虫为供试昆虫,用中华散尾鬼笔粗提物进行毒力测定,试验结果表明,中华散尾鬼笔粗提物对舞毒蛾3龄幼虫的校正死亡率为59。77%。
Optimization of Extracts from Lysurus mokusin Using Response Surface Methodology and Their Toxicity Test
Lysurus mokusin crude extract (LMCE) from Lysurus mokusin by ultrasonic method was used as an index in this test.The effects of different ultrasonic time,ethanol concentration,ultrasonic power and solid-liquid ratio on extracting rate of LMCE were investigated by single factor experiment,then the three factors,ultrasonic time,ultrasonic power and solid-liquid ratio,were chosen to determine the optimum process parameters in the response surface test.The results showed that under the conditions of 80 min of ultrasonic time,60% ethanol and 1 ∶ 80(g ∶ mL) of solid-liquid ratio,the extraction rate of LMCE was the highest (31.21%).The effects of LMCE on gypsy moth larvae were studied.The results showed that the adjusted mortality for the larvae of gypsy moth larvae was 59.77%.
