Bt水稻杂交后代中外源基因的遗传表达及抗性

以吉林省水稻主推品种"吉粳809"为母本,以Bt转基因水稻材料PP-121、PP-123及PP-136为父本进行杂交,系谱法筛选获得杂交后代材料Yj-276、Yj-231及Yj-288,通过外源基因的遗传表达及抗性研究,评价杂交后代育种价值。外源基因的遗传分析表明,cry1C和bar在杂交后代中连锁遗传并共分离;酶联免疫吸附剂(ELISA)分析表明,杂交后代中cry1C蛋白表达量正常,最高可达1。78 μg/g;抗性鉴定结果表明,杂交后代中二化螟抗虫性和除草剂Basta 1。0×10-3 mol/L耐性均>95%;农艺性状分析表明,部分杂交后代综合农艺性状优于杂交亲本"吉粳809",此结果为水稻抗虫育种提供新种质资源及理论依据。
Genetic Expression and Resistance Evaluation of Foreign Genes in Hybrid Lines of Bt Rice
In this study,Bt transgenic rice lines PP-121,PP-123 and PP-136 were crossed with Ji-jing 809,the main rice materials in Jilin province.The hybrid lines of Yj-276,Yj-231 and Yj-288 were obtained and the breeding value was evaluated through genetic expression and resistance evalu-ation of foreign genes.The results of genetic analysis showed that foreign genes of cry1C and bar were linked and co-segregated in the hybrid progenies.Enzyme Linked Immunosorbent Assay(ELISA)showed that cry1C protein was expressed normally in hybrid lines,up to 1.78 μg/g.Insect and herbicide resistance evaluation showed that three hybrid lines were highly resistant to striped stem borers and Basta 1.0 × 10-3 mol/L compared to female parents.Agronomic character analysis showed that some hybrid progenies had better performance than that of Jijing809.This study pro-vides new germplasm resources and a theoretical basis for insect-resistant rice breeding.
