以1年生球茎产生的原球茎为外植体,以MS为基本培养基,分别研究4种生长素和分裂素对原球茎增殖分化的影响以及激素组合对原球茎分枝状伸长增殖分化、原球茎分化产生球茎和球茎生根的影响。结果表明:NAA和ZT利于原球茎分枝状伸长增殖,在NAA 1。0mg/L+ZT 1。0mg/L时原球茎分枝状伸长增殖系数达到8。67;IAA和6-BA利于原球茎分化为球茎,在 IAA2。5 mg/L+6-BA 2。5 mg/L时原球茎分化为球茎系数达到5。14;IAA和6-BA利于原球茎分化产生的球茎生根,在IAA 2。5 mg/L+6-BA 0。1 mg/L时球茎产生根数达到6。26,每条根重达到9。90mg。
Rapid Propagation Technology of Seedlings from Protocorm of Oreor-chis patens(Lindl.)Lindl.
Using protocorm produced by one-year-old corm as explant and MS as basic culture me-dium,the effects of four auxins and mitogens on the proliferation and differentiation of protocorm were studied,and together studied were the effects of hormone combinations on the branching elonga-tion,proliferation and differentiation of protocorm,as well as the production of corm and rooting of corm during protocorm differentiation.The results showed that NAA and ZT were beneficial to the pro-liferation of protocorm by branching elongation,and the proliferation coefficient reached 8.67 at NAA 1.0 mg/L+ZT 1.0 mg/L.IAA and 6-B A were beneficial to the differentiation of protocorm into corm,and the differentiation coefficient reached 5.14 at IAA 2.5 mg/L+6-BA 2.5 mg/L.IAA and 6-BA were ben-eficial to the rooting of corm formation from protocorm,and the number of roots reached 6.26 per corm and the weight reached 9.90 mg per root at IAA 2.5 mg/L+6-BA 0.1 mg/L.
