摘要

通过对秀丽隐杆线虫各项生理指标及衰老相关基因的测定,观察红参水提物(RGWE)对线虫寿命的影响,并初步探索其作用机制.将秀丽隐杆线虫分别培养在普通培养基以及添加了不同质量浓度RGWE(0.8,1.4,2.0mg/mL)的培养基中,观察RGWE对线虫寿命、热应激、氧化应激、ROS含量、脂褐素含量的影响;通过qRT-PCR测定抗衰老相关基因的表达水平,初步探索RGWE抗衰老作用机制.结果表明:RGWE能极显著延长秀丽隐杆线虫的寿命,提高其抗应激能力,降低脂褐素水平及ROS含量,显著上调daf-16、sir2.1表达水平.

Abstract

Through the determination of various physiological indicators and aging related genes,the effect of water extract of red ginseng(RGWE)on the lifespan of Caenorhabditis elegans(C.elegans)and its possible mechanism were observed.C.elegans was cultured in the normal media containing different concentrations(0.8,1.4 and 2.0 mg/mL)of RGWE medium,and explored the effect of RGWE on life,heat stress,oxidative stress,ROS content and lipofuscin content.qRT-PCR was used to determine the expression level of anti-aging related genes,and the anti-aging mechanism of RGWE was initially explored.RGWE can significantly prolong the life-span of nematodes,improve anti-stress ability,reduce lipofuscin levels and ROS levels,and up-regulate significantly the expres-sion level of daf-16 and sir2.1 genes.
