Effects of Energy Supplement on Hepatic Metabolism and Injury of Perinatal Dairy Cows
Twelve components including eucommia ulmoides powder and rumen bypass glucose were mixed in a certain proportion.Six healthy cows and thirty cows with clinical ketosis were se-lected for toxicity experiment of energy supplement and therapeutic effect evaluation of ketosis in cows.The results showed that continuous administration of energy supplement for 7 days in healthy perinatal period did not induce the damage of liver and kidney function.After continuous administra-tion of energy supplement for 3 days in cows with ketosis,the concentration of β-hydroxybutyric acid and free fatty acid in blood was significantly decreased,while the concentration of glucose was signifi-cantly increased.The results of oxidative stress related indicators showed that administration of en-ergy supplement markedly increased the activities of peroxidase marker catalase(CAT)and glutathi-one peroxidase(GSH-PX)and decreased the malondialdehyde(MDA).The blood concentrations of in-flammatory cytokines IL-6,IL-8,and TNF-α were significantly decreased.In addition,it can im-prove the liver injury as evidenced by decreased activity of alanine aminotransferase(ALT),aspartate aminotransferase(AST),total bilirubin(TBIL)and indirect bilirubin(IBIL).Moreover,the phosphoryla-tion of hepatic energy metabolism and pathological injury regulator AMPK and SIRT3 protein con-tent were significantly increased.These results indicate that energy supplement administered by ga-vage can effectively relieve the negative energy balance,oxidative stress,inflammatory reaction,and liver injury of ketosis in dairy cows.
