Extraction Process of Total Alkaloids from Sinomenii caulis and Its Anti Gout Effect
Using sinomenine content measured by ultraviolet spectrophotometry as evaluation index,ethanol heating reflux extraction method was adopted to investigate the effects of ethanol concentra-tion,liquid-to-material ratio,extraction time and extraction times on the yield of Sinomenii caulis al-kaloids.The optimal extraction process parameters for extracting total alkaloids from Sinomenii cau-lis were analyzed using star point design-response surface method.Ethanol heating reflux and intra-peritoneal injection of potassium oxyzinate combined with intra-articular injection of sodium urate were used to create a rat model of gout.The effects of total alkaloids of Sinomenii caulis on the levels of UA,IL-1β,IL-6,TNF-α,XOD,BUN,Cr,Cys-c and ALT in serum and the levels of UA,IL-1β,IL-6 and TNF-α in synovial tissue of joint were detected by ELISA.HE staining was used to observe the protective effect of alkaloids from Sinomenii caulis on kidneys in rats with gout.The optimum ex-traction conditions of alkaloids from Sinomenii caulis obtained by star point design-response surface method were as follows:10 times the amount of 75%ethanol,reflux extraction 3 times,1.8 h each time;The total alkaloids of Sinomenii caulis can significantly improve the paw swelling caused by gout,reduce the levels of UA,IL-1β,IL-6,TNF-α,XOD,BUN,Cr,Cys-C and ALT in the serum of the model rats,and significantly reduce the contents of UA,IL-1β,IL-6 and TNF-α in the synovium of the rats.The results of HE staining showed that the total alkaloids can reduce kidney damage and inflammatory infiltration in rats with gout.The above research results indicate that star point design-response surface method is a simple and feasible method for optimizing the ethanol ex-traction process of total alkaloids from Sinomenii caulis.The established mathematical model has good predictive ability,and the total alkaloids from Sinomenii caulis have a significant anti gout ef-fect.
