Cloning and Bioinformatics Analysis of Lytic Polysaccharide Mono-oxygenase Gene PdLPMO9B from Pleurotus djamor
LPMO is a copper-containing lytic polysaccharide monooxygenase.The cDNA sequence of PdLPMO9B gene of Pleurotus djamor CM 13 strain was amplified by PCR technology,bioinfor-matics analysis was carried out,and finally a phylogenetic tree was constructed.The results show that PdLPMO9B gene has been successfully cloned.Its open reading frame ORF is 744 bp long,encoding 247 amino acids.The encoded protein has a molecular weight of 26.17 kDa,an isoelec-tric point of 5.91,and a signal peptide at the N-terminus,thus having strong hydrophobicity.The secondary structure is rich in β-sheet and random coil,and contains GH61 family domain and cel-lulose binding motif CBM.Three-dimensional modeling shows that it is closest to AA9 family mem-bers.The PdLPMO9B protein of Pleurotus djamor is closely related to GH61 protein encoded by Pleurotus ostreatus and clustered into one class.
