Bioinformatics Analysis of Trichoderma harzianum Cytochrome P450 Family Responding to Biological Stress Test of Bursaphelenchus xylophilus
Ten cytochrome P450(ThCYP)genes were selected from transcriptome data of Tricho-derma harzianum E15 under the stress test of Bursaphelenchus xylophilus,which were named ThCYP1~ThCYP10 in numerical order.The T.harzianum E15 ThCYP gene was studied by bioinfor-matics methods,such as gene physicochemical properties,phylogenetic relationship,amino acid se-quence alignment,gene structure analysis,conservative motif analysis and promoter analysis.These 10 ThCYP genes were basically similar in physicochemical properties,and the subcellular localiza-tion was observed mainly on the mitochondrial membrane.By establishing a phylogenetic tree of the CYP genes of Trichoderma virens and Trichoderma reesei and the E15 ThCYP gene of T.harzianum,it was found that the genetic relationship is very close.In amino acid multiple-sequences align-ments,these 10 ThCYP genes have multiple conserved sites to form conserved domains,including oxygen-binding domains,K-helix sequences,and conserved structural sequences of heme binding sites.In terms of gene structure,it was found that CDS regions are complete and the gene lengths are basically similar.Analysis of conserved motifs shows that the conserved motifs of the ten ThCYP genes are remarkably similar and share most of the same conserved motifs to play a key role in stress resistance.In the analysis of the promoter region,seven homeopathic components from the ten ThCYP genes are distributed in the promoter region,mainly ABRE and CGTCA-motifs.These prop-erties provide a remarkable contribution for resisting against B.xylophilus,where ThCYP2 has TC-rich repeats distributed in the promoter region,further increasing the resistance against B.xylophilus.
