Optimization of Protein Extraction from Pileus of Coprinus comatus and Analysis of Its Amino Acid
Ultrasonic assisted sodium hydroxide alkaline method was used to extract protein from pi-leus of Coprinus comatus.Through single factor test and orthogonal test,the effects of solid-liquid ra-tio,temperature,time,ultrasonic power,and pH value on protein extraction rate were analyzed,and the optimal extraction process was determined.The composition of amino acids was analyzed by pre-column derivatized high performance liquid chromatography,and the nutritional value of amino acids was preliminarily evaluated by amino acid ratio coefficient score(SRC)and chemical score(CS).The optimal extraction conditions were as follows:pH 11,50℃,solid-liquid ratio 1∶40,60 min,and ul-trasonic power 200 W.Under these conditions,the extraction rate was 33.18%.There are a variety of amino acids in the pileus of Coprinus comatus.The essential amino acids accounted for 35.9%of the total amino acids,and EAA/NEAA was 0.58,which is close to the WHO recommended value.
