新生期重复吸入七氟烷对成年大鼠海马体积与脑脊液Tau的影响

目的 探讨新生期大鼠多次吸入七氟烷后其成年期海马体积及脑脊液(cerebrospinal fluid,CSF)Tau蛋白的改变,确证磁共振成像(magnetic resonance imaging,MRI)扫描技术及临床上容易获取的标本CSF为间接推测七氟烷对认知功能影响的理论依据.方法 新生期SD大鼠 32 只,随机分成两组,即实验组(n=16),对照组(n=16).实验组于新生 7d(postnatal 7th day,P7)、新生 14d(postnatal 14th day,P14)、新生 21d(postnatal 21st day,P21)分别吸入浓度为 2.6%的七氟烷 2h,对照组于相同时间吸入对照混合运载气体(1L/min O2+1L/min Air)2h.自然喂养至成年期,于出生后第 96 天(postnatal 96th day,P96)使用MRI进行颅脑扫描,测量大鼠海马体积等相关指标,MRI扫描结束后,立即取CSF,标本收集完毕后按照ELISA试剂盒说明书要求测样本OD450mm 值,最后根据OD450mm 值算出Tau的浓度.结果 与对照组相比,实验组颅脑容积(brain volume,BV),左侧海马体积(left hippocampal volume,LHV),右侧海马体积(right hippocampal volume,RHV),最大颅脑长度(brain length,BL)均显著降低[对照组 BV,LHV,RHV,BL 分别为(2.19±0.10)cm3,(45.85±4.60)mm3,(46.04±2.90)mm3,(15.38±0.51)mm,实验组BV,LHV,RHV,BL分别为(1.88±0.10)cm3,(41.54±4.59)mm3,(43.12±2.70)mm3,(14.70±0.48)mm],差异具有统计学意义(t=-6.20,-1.88,-2.16,-2.83,P<0.05),实验组CSF中Tau蛋白浓度减小[对照组为(42.62±12.50)ng/mL,实验组为(28.93±6.13)ng/mL],有统计学差异(t=-2.23,P<0.05).结论 新生期大鼠重复多次吸入七氟烷麻醉后,CSF中Tau的总含量减少,但不能认为异常磷酸化Tau减少,其成年期海马体积减小,可认为MRI测量海马体积对认知功能障碍的早期诊断和预防具有一定临床指导意义.
Effects of neonatal repeated inhalation of sevoflurane on hippocampal volume and Tau in cerebrospinal fluid of adult rats
Objective To investigate changes of hippocampal volume and Tau in cerebrospinal fluid(CSF)of adult rats with neonatal repeated inhalation of sevoflurane,and to confirm magnetic resonance imaging(MRI)and CSF as the basis for inferring the effects of sevoflurane on cognitive function.Methods 32 neonatal SD rats were randomly divided into the experimental group(n=16)and the control group(n=16).Rats in experimental group inhalated 2.6%sevoflurane for 2 hours on postnatal 7th day(P7),P14 and P21 and rats in control group inhalated 1L/minO2+1L/min Air for 2 hours on the same days.After being naturally fed until adulthood,the rats underwent cranial MRI scanning on postnatal day 96(P96)to measure relevant indicators such as hippocampal volume.Immediately after the MRI scan-ning,cerebrospinal fluid(CSF)was collected.Following the completion of sample collection,the OD450mm values of the samples were measured according to the instructions of the ELISA kit.Finally,the Tau concentration was calculated based on the OD450mm values.Results Compared with control group,the brain volume(BV),left hippocampal volume(LHV),right hippocampal volume(RHV),brain length(BL)in experimental group were significantly lower(P<0.05)BV,LHV,RHV,and BL in control group were 2.19±0.10cm3,45.85±4.60mm3,46.04±2.90mm3,15.38±0.51mm,respectively;BV,LHV,RHV,and BL in experimental group were 1.88±0.10cm3,41.54±4.59mm3,43.12±2.70mm3,14.70±0.48mm,respectively,the differences were statvstically significant(t=-6.20,-1.88,-2.16,-2.83,P<0.05).The concentration of Tau protein in cerebrospinal fluid(CSF)was lower in the experimental group[control group:(42.62±12.50)ng/mL;experimental group:(28.93±6.13)ng/mL],with a statistical difference(t=-2.23,P<0.05).When comparing the other indicators between the groups,no statistically significant differences were found.Conclusions With neonatal repeated inhalation of sevoflurane,the total content of Tau in CSF of adult rats decreased,however,it can not be considered that the abnormal phosphorylated Tau decreased.The hippocampal volume decreased in adult,which can be considered that the MRI measurement of hippocampal volume has certain guiding significance for early diagnosis and prevention of cognitive impairment.
