LRP8 mediated Wnt/β-catenin signaling pathway promotes proliferation,migration and invasion of cervical cancer cells
Objective To explore the correlation between LRP8 expression in cervical cancer tissues and clinicopathological characteristics and prognosis,and to explore the effect and mechanism of LRP8 on the proliferation,migration and invasion of cervical cancer cells.Methods The expression of LRP8 in cervical cancer and adjacent tissues was detected by immunohistochemistry.According to the results of immunohistochemistry,30 cases of cervical cancer tissues were divided into LRP8 low expression group and LRP8 high expression group,and the correlation between LRP8 expression and age,pathological TNM stage,clinical stage and histological grade of cervical cancer patients was analyzed.The correlation between LRP8 expression and overall survival of cervical cancer patients was analyzed by GEPIA online database.LRP8 knockdown or overexpression plasmids were transfected into cervical cancer cells,and the LRP8 expression was detected by Western blot,the cell proliferation activity was detected by CCK-8,the cell migration and invasion ability was detected by Transwell,and the β-catenin expression was detected by Western blot.Results The expression of LRP8 was up-regulated in cervical cancer tissues.The high expression of LRP8 was associated with higher pathological T stage,pathological N stage,clinical stage and lower overall survival of cervical cancer patients,but not related to age,pathological M stage and histological grade.Knockdown of LRP8 reduced the proliferation,migration and invasion ability of cervical cancer cells and the expression level of β-catenin protein;conversely,overexpression of LRP8 increased the proliferation,migration and invasion ability of cervical cancer cells and the expression level of β-catenin protein.Conclusion LRP8 is highly expressed in cervical cancer tissues,and its high expression is associated with higher pathological T stage,pathological N stage,clinical stage and lower overall survival of cervical cancer patients.LRP8 may promote the proliferation,migration and invasion of cervical cancer cells by activating Wnt/β-catenin signaling pathway.
