BMP2 combined with chitosan hydrogel mediates VEGFA/ERK1/2 signaling pathway to regulate angiogenesis and promote tibial fracture healing in rats
Objective To explore the effect of BMP2 combined with chitosan hydrogel on tibial fracture healing in rats and its possible mechanism.Methods Thirty SD rats were divided into sham operation group,model group and BMP2 combined with chitosan hydrogel group,with 10 rats in each group.HE staining was used to detect the pathological changes at the fracture site.The serum ALP levels were detected by ELISA.The expression levels of Ang-1,CD31,VEGFA,VEGFR2 and p-ERK1/2 in rat bone tissue were detected by Western blot.Results Fracture healing was delayed in tibial fracture rats,and the serum ALP level and the expression levels of Ang-1,CD31,VEGFA,VEGFR2 and p-ERK1/2 in bone tissue of rats were lower than those in sham operation group.The injection of BMP2 combined with chitosan hydrogel promoted the fracture healing of tibial fracture rats,increased the serum ALP level and the expression levels of Ang-1,CD31,VEGFA,VEGFR2 and p-ERK1/2 protein in bone tissue of rats.Conclusion BMP2 combined with chitosan hydrogel induces angiogenesis and promotes fracture healing in tibial fracture rats,which may be related to its activation of VEGFA/ERK1/2 signaling pathway.
