Effect of LNC RNA Linc00152 on biological behavior and Fra-1/p53 gene of cervical cancer cells
Objective To explore the effect of LNC RNA Linc00152 on the biological behavior of cervical cancer cells and the Fra-1/p53 gene.Methods The expression of Linc00152 in human cervical cancer Hela cells and human normal cervical epithelial End1/E6E7 cells was detected by RT-PCR.Hela cells were divided into normal group(control group),transfection group(si-00152)and transfection control group(si-NC).The expression of Linc00152 was detected by RT-PCR.Proliferation of Hela cells was detected by MTT assay.Migration ability of Hela cells was detected by scratch test.Invasion ability of Hela cells was detected by Transwell method.The expressions of Fra-1 and p53 were detected by Western blot.Results The expression level of Linc00152 in Hela cells was significantly higher than that in End1/E6E7 cells(P<0.05).After transfection of si-Linc00152,The expression of Linc00152 was decreased significantly,proliferation,migration and invasion of Hela cells were significantly decreased(P<0.05),the expression of p53 was significantly increased,the expression of Fra-1 was significantly decreased(P<0.05).Conclusion Silencing the expression of LncRNA Linc00152 inhibited the proliferation,migration and invasion of Hela cells.
