姜黄素通过降低IL-18表达抑制人胰腺癌细胞的增殖和迁移

摘要
目的 探讨姜黄素对人胰腺癌细胞增殖、迁移和侵袭的影响及可能机制.方法 采用不同浓度姜黄素处理人胰腺癌细胞系SW1990,MTT法和克隆形成实验观察姜黄素对SW1990细胞增殖的影响;划痕试验和Tran-swell 试验观察姜黄素对SW1990细胞迁移和侵袭的影响;Real Time PCR和Western blot检测姜黄素对SW1990细胞IL-18和NF-κB基因和蛋白表达的影响.沉默IL-18后,观察SW1990细胞的增殖、迁移和侵袭能力变化.结果 姜黄素处理能抑制人胰腺癌细胞SW1990增殖,且呈剂量依赖性(P＜0.05);抑制人胰腺癌细胞SW1990的迁移和侵袭能力(P＜0.05);抑制人胰腺癌细胞SW1990中IL-18表达.沉默IL-18能降低人胰腺癌细胞增殖、迁移和侵袭能力,且下调IL-18引起的效应与姜黄素的效果一致.结论 姜黄素可以明显抑制人胰腺癌细胞增殖、迁移和侵袭,其抗肿瘤作用与降低IL-18表达密切相关.
Abstract
Objective To explore the effect of curcumin on proliferation,migration and invasion of human pancreatic cancer cells and its possible mechanism.Methods Human pancreatic cancer cell line SW1990 was treated with different concentrations of curcumin,and the effects of curcumin on the proliferation of SW1990 cells were observed by MTT method and clone formation experiment.Scratch test and Transwell assay were used to observe the effects of curcumin on the migration and invasion of SW1990 cells.Real Time PCR and Western blot were used to detect the effects of curcumin on the expression of IL-18 and NF-κB mRNA and proteins in SW1990 cells.After silencing IL-18,the proliferation,migration,and invasion ability of SW1990 cells was observed.Results Curcumin treatment could inhibit the proliferation of human pancreatic cancer cell SW1990 in a dose-dependent manner(P＜0.05).Curcumin inhibited the migration and invasion of human pancreatic cancer cell SW1990(P＜0.05).Curcumin inhibits the expression of IL-18 in human pancreatic cancer cell SW1990.Silencing IL-18 can reduce the proliferation,migration and invasion of human pancreatic cancer cells,and the effect of down-regulation of IL-18 is consistent with that of curcumin.Conclusion Curcumin can significantly inhibit the proliferation,migration and invasion of human pancreatic cancer cells,and its anti-tumor effect is closely related to the reduction of IL-18 expression.
