补肾活血方联合非布司他对高尿酸性肾病大鼠炎症损伤的影响及可能机制

目的 探讨补肾活血方联合非布司他对高尿酸性肾病大鼠炎症反应和肾损伤的改善作用及可能机制.方法 雄性Sprague-Dawley大鼠随机分为对照组(Control)、模型组(Model)、补肾活血汤组(BSHXDS)、非布司他组(Febuxostat)、补肾活血汤联合非布司他组(BSHXDS+Febuxostat),每组12只.生化分析检测各组大鼠血清尿酸和肌酐水平;HE染色检测各组大鼠肾脏组织损伤;ELISA检测各组大鼠血清IL-6和IL-1β水平;Western blot检测肾组织TLR4、MyD88、NF-kappa B p65蛋白的表达.结果 补肾活血汤、非布司他和补肾活血方联合非布司他均能降低高尿酸性肾病大鼠肾组织炎症性损伤、血清尿酸、肌酐、炎症因子水平,同时下调大鼠肾组织TLR4、MyD88、NF-kappa B的表达水平;与单独使用补肾活血汤或非布司他相比,补肾活血汤联合非布司他组的作用更为明显.结论 补肾活血方联合非布司他可通过调控TLR4/MyD88/NF-kappa B通路抑制高尿酸性肾病大鼠炎症反应,改善肾功能.
Effect and possible mechanism of Bushen Huoxue decoction combined with febuxostat on inflammation injury in rats with hyperuricemic nephropathy
Objective To investigate the effect and possible mechanism of Bushen Huoxue decoction combined with febuxostat on inflammation and renal injury in rats with hyperuricemic nephropathy.Methods Male Sprague-Dawley rats were randomly divided into control group,model group,Bushen Huoxue decoction group(BSHXDS),Febuxostat group,Bushen Huoxue decoction combined Febuxostat group(BSHXDS+Febuxostat),with 12 rats in each group.The levels of Uric acid and creatinine in serum were detected by biochemical analysis.Renal tissue injury was observed by HE staining.The levels of IL-6 and IL-1β in serum were detected by ELISA.The expressions of TLR4,MyD88 and NF-kappa B p65 in renal tissues were detected by Western blot analysis.Results Bushen Huoxu decoction,febuxositat and Bushen Huoxu decoction combined with Febuxositat could reduce the inflammatory injury,the levels of uric acid and creatinine in serum,and inflammatory factors in renal tissue of rats with hyperuricemic nephropathy,and down-regulate the expressions of TLR4,MyD88 and NF-kappa B in renal tissue of rats.The effect of Bushen Huoxue decoction combined with febuxostat group was more obvious than that of Bushen Huoxue decoction or febuxostat alone.Conclusion The Bushen Huoxue decoction combined with febuxostat could inhibit the inflammatory response and improve renal function in rats with hyperuric acid nephropathy by regulating TLR4/MyD88/NF-kappa B pathway.
