Effect of platelet lysate on the expression of Runx2 and SOX9 in cartilage injury of Wistar rats
Objective To investigate the effect of platelet lysate(PL)on the expressions of Runx2 and SOX9 in the repair of traumatic cartilage injury in Wistar rats.Methods Thirty Wistar rats were randomly divided into control group,saline group and platelet lysate group,with 10 rats in each group.HE staining and safranin O-fast green staining were used to observe the knee cartilage of rats in each group,and Mankin score was used.Immunohistochemical staining was used to detect the expressions of Runx2 and SOX9 in knee cartilage of rats,and Western blot was used to detect the expressions of Runx2 and SOX9 in knee cartilage of rats.Results After PL injection,the injury of cartilage tissue in rats with traumatic knee cartilage injury was significantly alleviated,the Mankin score of cartilage was significantly reduced,the number of cells of Runx2 and SOX9 positive expression in knee cartilage tissue was significantly reduced,and the protein expressions of Runx2 and SOX9 in knee cartilage tissue were significantly reduced(P<0.01).Conclusion Platelet lysate can promote the structural recovery of cartilage tissue in Wistar rats and reduce the expression of Runx2 and SOX9 after cartilage injury.
