ATP4A combined with ATP4B can be used as a biomarker for early diagnosis and prognosis of gastric cancer
Objective To screen potential biomarkers for early diagnosis and prognosis of gastric cancer by bioinformatics. Methods GEO2R was used to screen differentially expressed genes from GEO data set,and GO/KEGG was used for functional enrichment analysis. The STRING tool was used to generate the protein interaction network, and the hub genes were identified using the MCODE plugin of Cytoscape. The expression of Hub gene in gastric cancer tissues was analyzed using HPA,and the prognosis and survival curve was analyzed based on Kaplan-Meier.Results 105 intersecting DEGs were obtained from three datasets GSE13911,GSE19826 and GSE29272,which were mainly involved in metabolic pathways.A total of 75 protein interaction nodes were screened, and 10 Hub genes were obtained, among which ATP4A and ATP4B were significantly correlated(r=0.699,P<0.001).The expressions of ATP4A and ATP4B in the cytoplasm of gastric cancer tissue were increased, and were positively correlated with poor prognosis. Conclusion ATP4A and ATP4B may be potential biomarkers for early diagnosis and prognosis of gastric cancer.
