Experimental study of FERMT1 affecting the proliferation,migration,invasion and apoptosis of prostate cancer cells by regulating the NF-κB signaling pathway
Objective To explore the effect of FERMT1 on the proliferation,migration,invasion and apoptosis of prostate cancer cells and its regulation of NF-κB signaling pathway.Methods qRT-PCR and Western blot were used to detect the expression of FERMT1 in normal prostate epithelial cells P69 and prostate cancer cells LNCaP,DU145,and PC-3.si-NC,si-FERMT1,pcDNA-NC,and pcDNA-FERMT1 were transfected into LNCaP cells.After si-FERMT1 was transfected into LNCaP cells,NF-κB activation activator TNF-α was added to treat the cells.MTT test was used to detect cell viability.Flow cytometry was used to detect the apoptosis rate.The transwell assay was used to detect cell migration and invasion.Western blot was used to detect the expressions of CyclinD1,MMP2,MMP9,Cleaved-caspase-3,p-p65,and p-IκBα protein.Results The expression of FERMT1 was increased in prostate cancer cells.Interfering with the expression of FERMT1 could inhibit cell proliferation,migration,invasion and the expressions of CyclinD1,MMP2,MMP9,p-p65,p-IκBα,and promote cell apoptosis and the expression of Cleaved-caspase-3.Overexpression of FERMT1 could promote cell proliferation,migration,invasion and the expressions of CyclinD1,MMP2,MMP9,p-p65,p-IKBα,and inhibit cell apoptosis and the expression of Cleaved-caspase-3.NF-κB activation activator TNF-α could reverse the effect of interfering with the expression of FERMT1 on the biological behavior of cells after treating the cells.Conclusion Interfering with the expression of FERMT1 could reduce the proliferation,migration and invasion of prostate cancer cells,and promote cell apoptosis,its mechanism of action was related to the inhibition of the activation of the NF-κB signaling pathway.
