The effect and molecular mechanism of miR-129-5p targeting KLK7 on the proliferation and invasion of ovarian cancer SKOV3 cells
Objective To explore the effects and mechanisms of high expression of miR-129-5p on the proliferation and invasion of ovarian cancer SKOV3 cells.Methods qRT-PCR was used to detecte the expression of miR-129-5p in SKOV3 cells and IOSE80 cells.miR-129-5p mimics was transfected into SKOV3 cells,the expression of miR-129-5p and KLK7 in each group were detected by qRT-PCR,the expression of KLK7 was detected by Western blot,the targeting relationship between KLK7 and miR-129-5p was analyzed by targetscan 7.2 and double luciferase experiment,the proliferation and invasion of cells was detected by CCK8 and Transwell.Results The expression of miR-129-5p in SKOV3 cells was lower than that in IOSE80 cells(P<0.05).Over expression of miR-129-5p decreased the expression of KLK7 in SKOV3 cells(P<0.05).Targetscan 7.2 analysis and double luciferase experiment confirmed that KLK7 was the target gene of miR-129-5p.Over expression of miR-129-5p could inhibit the proliferation and invasion of SKOV3 cells(P<0.05).Conclusion Overexpression of miR-129-5p could inhibit the proliferation and invasion of ovarian cancer SKOV3 cells by targeting KLK7.
