目的 探讨高表达miR-129-5p对卵巢癌SKOV3细胞增殖和侵袭的影响及分子机制.方法 采用实时荧光定量PCR方法检测卵巢癌SKOV3细胞与人正常卵巢上皮细胞株中miR-129-5p的表达.在卵巢癌SKOV3细胞中瞬时转染miR-129-5p模拟物及无意义序列,采用CCK8法及Transwell实验检测各组细胞的增殖能力和侵袭能力;实时荧光定量PCR检测miR-129-5p和KLK7的相对表达水平;Western blot检测各组细胞中KLK7蛋白表达;Targetscan 7.2和双荧光素酶实验分析miR-129-5p与KLK7的靶向关系.结果 miR-129-5p在卵巢癌SKOV3细胞中的表达低于人正常卵巢上皮细胞(P<0.05),过表达miR-129-5p降低SKOV3细胞中KLK7 mRNA和蛋白的表达,Targetscan 7.2网站分析及双荧光素酶实验证实KLK7为miR-129-5p的靶基因,过表达miR-129-5p抑制SK-OV3细胞增殖和侵袭能力(P<0.05).结论 过表达miR-129-5p可通过靶向调控KLK7抑制卵巢癌SKOV3细胞增殖和侵袭.
The effect and molecular mechanism of miR-129-5p targeting KLK7 on the proliferation and invasion of ovarian cancer SKOV3 cells
Objective To explore the effects and mechanisms of high expression of miR-129-5p on the proliferation and invasion of ovarian cancer SKOV3 cells.Methods qRT-PCR was used to detecte the expression of miR-129-5p in SKOV3 cells and IOSE80 cells.miR-129-5p mimics was transfected into SKOV3 cells,the expression of miR-129-5p and KLK7 in each group were detected by qRT-PCR,the expression of KLK7 was detected by Western blot,the targeting relationship between KLK7 and miR-129-5p was analyzed by targetscan 7.2 and double luciferase experiment,the proliferation and invasion of cells was detected by CCK8 and Transwell.Results The expression of miR-129-5p in SKOV3 cells was lower than that in IOSE80 cells(P<0.05).Over expression of miR-129-5p decreased the expression of KLK7 in SKOV3 cells(P<0.05).Targetscan 7.2 analysis and double luciferase experiment confirmed that KLK7 was the target gene of miR-129-5p.Over expression of miR-129-5p could inhibit the proliferation and invasion of SKOV3 cells(P<0.05).Conclusion Overexpression of miR-129-5p could inhibit the proliferation and invasion of ovarian cancer SKOV3 cells by targeting KLK7.
