TPD52 regulates apoptosis and cell cycle of breast cancer cells by regulating PI3K/AKT signaling pathway
Objective To investigate the expression level of TPD52 in breast cancer and its effect on cell apoptosis and cell cycle.Methods GEPIA database was used to analyze the expression of TPD52 in breast cancer tissues and normal tissues.Kaplan Meier plotter was used to analyze the relationship between TPD52 expression and the prognosis of breast cancer patients.Human breast cancer MDA-MB-453 and HCC1937 cell lines were cultured in vitro.TPD52 small interfering RNA was transfected into MDA-MB-453 and HCC1937 cells to knockdown the expression of TPD52.qRT-PCR was used to detect the expression of TPD52 mRNA.Flow cytometry was used to detect apoptosis rate and cell cycle of breast cancer cells.Western blot was used to detect the expression of p-PI3K and p-Akt in breast cancer cells.Results GEPIA database showed that the expression level of TPD52 in breast cancer tissues was significantly higher than that in normal breast tissues.High expression of TPD52 was significantly correlated with poor prognosis in breast cancer patients.Knockdown of TPD52 significantly promoted apoptosis and arrested cell cycle progression of breast cancer cells.In addition,knockdown of TPD52 inhibited the expression of p-PI3K and p-Akt in breast cancer cell lines.Conclusion TPD52 is highly expressed in breast cancer tissues,which can promote apoptosis and arrest cell cycle progression by regulating PI3K/AKT signaling pathway.
