目的 研究乳腺癌中TPD52的表达水平及对人乳腺癌细胞凋亡和细胞周期的影响.方法 通过GE-PIA数据库分析乳腺癌组织中TPD52表达情况;利用Kaplan Meier plotter数据库分析TPD52的表达和乳腺癌患者预后的关系;体外培养人乳腺癌MDA-MB-453和HCC1937细胞系,将TPD52小干扰RNA分别转染进MDA-MB-453和HCC1937细胞系中敲低TPD52的表达.qRT-PCR检测乳腺癌细胞系中TPD52 mRNA的表达;流式细胞术检测乳腺癌细胞凋亡率和细胞周期.Western blot实验检测乳腺癌细胞中p-PI3K和p-AKT蛋白的表达.结果 GEPIA数据库显示TPD52在乳腺癌组织中表达水平显著高于正常乳腺组织;TPD52高表达和乳腺癌患者预后较差显著相关;敲低TPD52可显著促进乳腺癌细胞凋亡并阻滞细胞周期进程、抑制乳腺癌细胞系中p-PI3K和p-AKT蛋白的表达.结论 TPD52在乳腺癌组织中高表达,其可通过调控PI3K/AKT信号通路促进乳腺癌细胞凋亡并阻滞细胞周期进程.
TPD52 regulates apoptosis and cell cycle of breast cancer cells by regulating PI3K/AKT signaling pathway
Objective To investigate the expression level of TPD52 in breast cancer and its effect on cell apoptosis and cell cycle.Methods GEPIA database was used to analyze the expression of TPD52 in breast cancer tissues and normal tissues.Kaplan Meier plotter was used to analyze the relationship between TPD52 expression and the prognosis of breast cancer patients.Human breast cancer MDA-MB-453 and HCC1937 cell lines were cultured in vitro.TPD52 small interfering RNA was transfected into MDA-MB-453 and HCC1937 cells to knockdown the expression of TPD52.qRT-PCR was used to detect the expression of TPD52 mRNA.Flow cytometry was used to detect apoptosis rate and cell cycle of breast cancer cells.Western blot was used to detect the expression of p-PI3K and p-Akt in breast cancer cells.Results GEPIA database showed that the expression level of TPD52 in breast cancer tissues was significantly higher than that in normal breast tissues.High expression of TPD52 was significantly correlated with poor prognosis in breast cancer patients.Knockdown of TPD52 significantly promoted apoptosis and arrested cell cycle progression of breast cancer cells.In addition,knockdown of TPD52 inhibited the expression of p-PI3K and p-Akt in breast cancer cell lines.Conclusion TPD52 is highly expressed in breast cancer tissues,which can promote apoptosis and arrest cell cycle progression by regulating PI3K/AKT signaling pathway.
