microRNA-195-5p inhibits the proliferation and metastasis of trophoblast cells by regulating LRP6
Objective To investigate the effect and possible mechanism of miRNA-195-5p on proliferation,migration and invasion of trophoblast cells(HTR-8/SVneo).Methods Trophoblast cells(hTR-8/SVneo)were cultured in vitro,and miR-195-5p mimics were transfected into trophoblast cells(hTR-8/SVneo)to increase the expression of miR-195-5p.The expression of miR-195-5p and LRP6 mRNA were detected by real-time quantitative PCR in each group.The proliferation of HTR-8/SVneo cells were detected by CCK-8 assay,and the migration and invasion of HTR-8/SVneo cells were detected by Transwell assay.Results miR-195-5p mimics significantly promoted the expression of miR-195-5p and inhibited the expression of LRP6 mRNA in HTR-8/SVneo cells(P<0.05).Dual luciferase assay showed that miR-195-5p could directly target LRP6.After transfection with miR-195-5p mimics,the proliferation,migration and invasion abilities of HTR-8/SVneo cells were decreased.Conclusion miR-195-5p may inhibit the proliferation,migration and invasion of HTR-8/SVneo cells by inhibiting the expression of LRP6.
