Dexmedetomidine improve cognitive function in elderly rats after femoral neck fracture
Objective To investigate the mechanism of dexmedetomidine to improve cognitive function after femoral neck fracture in elderly rats.Methods Thirty rats were randomly divided into sham group,model group and dexmedetomidine group.The left femoral neck fracture model of aged rats was established.The cognitive function of rats was observed by morris water maze.The pathological injury of hippocampus was observed by HE staining.The number of neurons in hippocampus region was observed by Nissl staining.The positive expressions of Iba-1/iNOS and Iba-1/Arg-1 were detected by immunofluorescence staining.The expressions of IL-1 β and IL-18 in brain of rats were determined by ELISA kit.The expressions of TXNIP,NLRP3,Caspase-1 and ASC in brain of rats were detected by Western blot.Results Dexmedetomidine could improve the cognitive function of rats,reduce the pathological injury of hippocampus,increase the number of neurons,decrease the number of Iba-1/iNOS positive cells,and increase the number of Iba-1/Arg-1 positive cells,decrease the expressions of IL-1β and IL-18,TXNIP,NLRP3,Caspase-1 and ASC in brain tissue.Conclusion Dexmedetomidine can improve cognitive function after femoral neck fracture in elderly rats by regulating microglia polarization,and its mechanism may be related to the regulation of TXNIP/NLRP3 signaling pathway.
