Crocetin inhibits the malignant development of colon cancer cells through PI3K/Akt pathway
Objective To investigate whether crocetin can inhibit the malignant development of colon cancer cells through PI3K/Akt pathway.Methods Colon cancer HT29 and SW480 cell lines were treated with crocetin and divided into control group(untreated HT29 and SW480 cells,NC group)and crocetin group(HT29 and SW480 cells treated with crocetin,Cro group).The effects of crocetin on proliferation,apoptosis,migration and invasion of HT29 and SW480 cells were determined by plate cloning assay,Annexin V-FITC/PI double staining assay,scrape assay and Transwell assay.The expressions of PI3K,p-PI3K,Akt and p-Akt were detected by Western blot.Results Compared with the control group,after treatment with crocetin,the proliferation,migration and invasion ability of HT29 and SW480 cells were significantly decreased,the apoptosis rate was significantly increased,and the expressions of p-PI3K and p-Akt protein in colon cancer cells was significantly decreased(P<0.05).Conclusion Crocetin can inhibit the proliferation,migration and invasion of colon cancer cells and promote apoptosis by inhibiting PI3K/Akt pathway.
