Effect and mechanism of ethanol extract from radix of actinidia chinensis on proliferation,apoptosis and invasion of Hela cells
Objective To investigate the effect and its molecular mechanism of ethanol extract from radix of actinidia chinensis(EERAC)on proliferation,apoptosis and migration of Hela cells.Methods Human cervical cancer Hela cells were cultured in vitro,treated with 25,50,100 µg/mL of EERAC.After 48h,the culture was terminated,and the proliferation of Hela cells was detected by MTT method.The apoptosis of Hela cells was detected by flow cytometry.The invasion of Hela cells was detected by transwell assay.The expressions of Cyclin 1(Cyclin D1),cyclin dependent kinase inhibitor(P21)and matrix metalloproteinase-2(MMP-2)were detected by Western blot.Results After treatment with different concentrations of EERAC,the proliferation of Hela cells was decreased,the apoptosis rate was increased,the ability of invasion of Hela cells was decreased,and the expressions of CyclinD1 and MMP-2 were decreased,while the expression of P21 was increased in a concentration-dependent manner.Conclusions EERAC could inhibit the proliferation and invasion of Hela cells and induce apoptosis.
