摘要

目的 本研究旨在探讨miR-365b-3p调控胶质瘤血管生成拟态的潜在机制.方法 应用实时定量PCR检测miR-365b-3p在胶质瘤细胞系U87细胞中的表达水平;应用双荧光素酶报告基因分析系统检测miR-365b-3p和FLI1的结合作用;应用CCK-8法、Transwell法、体外管形成实验检测miR-365b-3p或FLI1表达变化对U87细胞增殖、迁移、侵袭和管形成能力的影响.结果 miR-365b-3p在U87细胞中低表达,过表达miR-365b-3p显著抑制胶质瘤细胞的增殖、迁移、侵袭及管形成能力.FLI1在U87细胞中高表达,沉默FLI1抑制胶质瘤细胞的管形成能力.miR-365b-3p通过靶向结合FLI13'UTR,抑制其mRNA和蛋白的表达.结论 miR-365b-3p通过靶向结合FLI1抑制胶质瘤细胞血管拟态生成.

Abstract

Objective To investigate the potential mechanism by which miR-365b-3p regulates vasculogenic mimicry in gliomas.Methods Quantitative real-time PCR was used to detect the expression level of miR-365b-3p in U87 cells.The binding of miR-365b-3p and FLI1 was detected by luciferase reporter assay.The effects of miR-365b-3p was FLI1 expression on the proliferation,migration,invasion and formation ability of VM in U87 cells were examined by CCK-8,transwell and in vitro tube formation assays.Results miR-365b-3p is low-expressed in U87 cells,and overexpression of miR-365b-3p significantly inhibits the proliferation,migration,invasion,and tube formation of glioma cells.FLI1 was highly expressed,down-regulation of FLI1 inhibited the tube-forming ability of glioma cells.miR-365b-3p inhibited expression of FLI1 mRNA and protein by targeting its 3'UTR.Conclusion miR-365b-3p inhibits vasculogenic mimicry of glioma cells via targeting FLI1.
