The mechanism of Hedyotisdiffusa-Scutellariabarbata combined with cisplatin in the treatment of lung cancer
Objective To explore the mechanism of Hedyotisdiffusa(HD)-Scutellariabarbata(SBD)combined with cisplatin in the treatment of lung cancer.Methods The human non-small cell lung cancer cell line A549 was selected,and the cells were given low,medium and high concentrations of HD-SBD(20,40,60 mg/L)and low,medium and high concentrations of cisplatin(5,10,20 mg/L).The best concentrations of HD-SBD and cisplatin were screened by CCK8.The follow-up experimental cells were divided into control group,HD-SBD group,cisplatin group,and combined group.Flow cytometry was used to detect apoptosis of cells.Transwell assay was used to detect invasion of cells.The migration of cells was tested by wound healing assay.The level of EMT and TGF-β/Smad pathway related proteins was detected bywestern blot.Results The optimal concentrations for HD-SBD and cisplatin were 60 mg/L and 20 mg/L,respectively.Both HD-SBD and cisplatin treated separately reduced apoptosis rate,invasion and migration rate of cells,and inhibited EMT process and TGF-β/Smad signaling pathway.The combined treatment of HD-SBD and cisplatin further reduced apoptosis,migration and invasion of cells,and further inhibited the EMT process and TGF-β/Smad signaling pathway.Conclusion HD-SBD combined with cisplatin promoted the apoptosis of lung cancer cells and inhibits the migration,invasion and EMT process of lung cancer cell migration by regulating TGF-β/Smad signaling pathway.
