Yangyin Huowei mixture improving chronic atrophic gastritis in rats through NLRP3/Caspase-1/GSDMD signaling pathway
Objective To explore the mechanism of Yangyin Huowei mixture in treating chronic atrophic gastritis(CAG)based on NLRP3/Caspase-1/GSDMD signaling pathway.Methods Forty SD rats were randomly divided into control group,model group,low dose and high dose of Yangyin Huowei mixture group.The CAG model of rats was reproduced by comprehensive method(N-Methyl-N'-nitro-N-nitrosoguanidine+ranitidine+ethanol with different concentration+starvation and satiety disorder).The general state and body weight of rats were observed.Enzyme-linked immunosorbent assay(ELISA)was used to detect the levels of pepsinogenⅠ(PGI),pepsinogen Ⅱ(PG Ⅱ)),interleukin-1β(IL-1β),IL-6,IL-18 levels in serum of rats.The pathological injury of gastric mucosa was observed by HE staining.The expressions of NLRP3,Caspase-1,GSDMD and GSDMD-N in gastric tissues were detected by Western blot.Results Compared with model group,low and high doses of Yangyin Huowei mixture could significantly increase the acidity of gastric juice,improve the pH value of gastric juice,significantly increase the levels of PGI and PGR,and decrease the level of PG Ⅱ,reduce inflammation in gastric mucosal,decrease the levels of IL-1β,IL-6 and IL-18 in serum,decrease the levels of NLRP3,Caspase-1,GSDMD and GSDMD-N in gastric tissue of rats.Conclusion Yangyin Huowei mixture improves gastric mucosal inflammation in CAG rats,which may be achieved by regulating NLRP3/Caspase-1/GSDMD signaling pathway to inhibit pyroptosis.
