CircSPECC1 promotes cervical cancer cell proliferation and migration through the miR-409/SHP-2 axis
Objective To explore the effect and mechanism of circSPECC1 on the proliferation and migration of cervical cancer cells.Methods The co-localization of circSPECC1 and miR-409-3p in cervical cancer cells was observed by fluorescence in situ hybridization.Dual luciferase reporter gene assay confirmed circSPECC1 binding to miR-409-3p.The effect of overexpression of miR-409-3p on the clonogenesis of cirCSPECC1-overexpressed cervical cancer cells was detected by plate cloning assay.The effect of overexpression of miR-409-3p on the migration ability of cirCSPECC1-overexpressed cervical cancer cells was detected by transwell assay.The binding between SHP-2 mRNA and miR-409-3p was verified by double luciferase reporter gene assay.The effects of overexpression or inhibition of miR-409-3p on the expression of SHP-2 protein in cervical cancer cells were detected by Western blot.Results CircSPECC1 and miR-409-3p was co-localized in the cytoplasm of cervical cancer cells.CircSPECC1 binded to miR-409-3p,and overexpression of miR-409-3p reversed the promoting effect of overexpression of circSPECC1 on the cloning and migration ability of cervical cancer cells.SHP-2 mRNA binded to miR-409-3p.Inhibition of miR-409-3p upregulated the expression of SHP-2 in cervical cancer cells and overexpression of miR-409-3p reduced the expression of SHP-2 in cervical cancer cells.Conclusion CircSPECC1 promotes cervical cancer cell proliferation and migration by binding to miR-409-3p,which may be related to the upregulation of SHP-2 expression by sponge miR-409-3p.
