The role and mechanism of miR-622 in osteoarthritis
Objective To analyze the role and possible mechanism of microRNA-622(miR-622)in osteoarthritis.Methods Human chondrocytes C28/I2 were cultured in vitro and divided into control group,down-regulated miR-622 group and up-regulated miR-622 group.The proliferation of cells in each group was detected by MTT method.The apoptosis of cells in each group was detected by flow cytometry.The levels of TNF-α,IL-1 β and MMP-13 were detected by ELISA.Dual luciferase reporting experiment verified the targeting relationship between miR-622 and SDF-1 and CXCR4.The expressions of SDF-1 and CXCR4 mRNA otein were detected by RT PCR and Western blot.Results Compared with the control group,after down-regulating the expression of miR-622,the proliferation rate of C28/I2 cells was decreased,and the apoptosis rate,TNF-α,IL-1β and MMP-13 expression levels were increased.After the up-regulation of miR-622 expression,the proliferation rate of C28/I2 cells was increased,and the apoptosis rate,TNF-α,IL-1β and MMP-13 expression levels were decreased.Dual luciferase reports confirmed that SDF-1 and CXCR4 were the target genes of miR-622.Compared with the control group,after down-regulating the expression of miR-622,the expressions of SDF-1 and CXCR4 mRNA and protein in C28/I2 cells were up-regulated.After the up-regulation of miR-622 expression,the expressions of SDF-1 and CXCR4 mRNA and protein in C28/I2 cells were decreased.Conclusion miR-622 was involved in the pathogenesis of osteoarthritis by regulating the SDF-1/CXCR4 axis.
